Flow cytometry

Flow cytometry is a high-output technique that allows quantitative multiparametric analysis (light scattering measurments, fluorescence properties) at the single-cell level.

Equipment:

  • Tecnai 12 trasnmission electron microscope equipped with a CCD camera (OneView 4Kx4K Gatan) driven by the GMS software (self-service after training)
  • TeneoVS scanning electron microscope, equipped with ETD, SE and BSE detectors and the VolumeScope module (for Serial-Block-Face mode) driven by FEI and Maps2 software (self-service after training)
  • Fischione 2020 tomography object holder, with tilt up to +/-80°.
  • Leica EM PACT2 high pressure cryofixation unit
  • Leica AFS devices: automated cryosubstitution.
  • ultramicrotomes (1 Leica UC6, 1 Leica UCT): production of semi- and ultra-fine sections of biological samples embedded in various resins (self-service after training).
  • Leica UC7-FC7 ultracryomicrotome: making semi- and ultra-fine sections of frozen biological samples.
  • Cressington 308R carbon evaporator: vacuum evaporation of carbon on grids (self-service after training).

Image processing and analysis

 

The Platform provides help and assistance to users for image processing, analysis, quantification and interpretation.

Matlab (MathWorks) : Digital calculation programming software allowing the development of advanced processing (image processing, signal processing, statistics, etc.) on images. It can be interfaced with the software Imaris.

Huygens Pro (SVI – Scientific Volume Imaging) : image deconvolution software; it allows the deflowering and denoising of images based on the knowledge of the physical and optical characteristics of the microscopes and the sample.

ImageJ et Fiji: free software for processing and analysing 3-4-5 D images with numerous plugins and allowing the creation of custom scripts (macros).

The software is installed on 2 powerful workstations.

A graphics tablet is also available, facilitating manual image segmentation operations.
