Deciphering the dynamic molecular architecture of centrioles and meiotic chromosomes

“Uncovering the architecture of complex, multi-protein containing macromolecular assemblies such as chromosomes or centrioles is key to understand the mechanism of their biogenesis and modes of function. To answerer open questions concerning organizational principles, assembly and disassembly pathways of centrioles and chromosomes, we apply various super-resolution techniques (e.g. SIM, STED and Cryo-ET) in combination with specific cytological methods (e.g. molecular standards, expansion microscopy and whole-gonad/embryo spreading).

In this seminar I will present some of our published and unpublished findings concerning chromosome cohesion and DNA loop organization during meiotic prophase of C. elegans and how population derived data (from e.g. ChIP-Seq or Hi-C analyses) compare to how a given chromosome is organized in a given moment in time in worms and yeast. In the second part, I will share our published and unpublished results about the molecular architecture and unusual cartwheel organization of C. elegans centrioles and the mechanism of centriole elimination during oogenesis of worms and in Naegleria gruberi during its flagellate-to-amoeba conversion.”
