Starting from the end: mechanism and regulation of mRNA decay
We are interested to unravel the mechanisms controlling eukaryotic gene expression at the level of RNA decay. Changes in mRNA degradation contribute to the regulation of gene expression by affecting the synthesis of specific proteins through the modulation of the levels of the corresponding mRNA templates. But it also participates to other process such as maintaining the overall cellular mRNA concentration or facilitating localized expression of specific factors. Besides its quantitative role in controlling mRNA levels, RNA decay also performs a second qualitative function through the elimination of aberrant or damaged RNA molecules.
After quick review of the basics of mRNA decay, I will provide some new insights into this process recently gained in our lab from the studies in yeast and mammals. This will include regulation of poly(A) tail shortening, mechanisms impacting cleavage of the mRNA cap, and the role of the m6A RNA epitranscriptomic mark during yeast meiosis. In some instances, defects into the function of the corresponding factors in human have been linked to diseases.
