Building to understand: Enhancer-Promoter Communication in the Context of Regulatory Landscapes

Résumé :

Enhancers can regulate promoters from a distance, but it is still unclear how they establish this  communication. Furthermore, multiple enhancers often control the activity of the same gene, yet their interactions remain elusive. We have established a synthetic framework to build regulatory landscapes within the genome of mouse embryonic stem cells to dissect enhancer promoter communication. We have now expanded our toolbox to test enhancer libraries, alone or in combination with other elements, to identify principles of distal gene regulation.
