DESCRIPTION:Invited by the Ribes/Nedelec\, Olivier Pourquié (Frank Burr Mallory Professor of Pathology\, Brigham and Women’s Hospital; Professor of Genetics\, Harvard Medical School) will present an Institut Jacques Monod seminar on the theme: \nDeconstructing human musculo-skeletal development in vitro  \nAbstract: \nSkeletal muscles and vertebrae derive from precursors located in the embryonic segments called somites. These structures form periodically from a posterior tissue called Presomitic Mesoderm (PSM). The rhythmic formation of somites involves a molecular oscillator called segmentation clock which drives pulses of Notch\, Wnt and FGF signaling in the PSM. Virtually nothing is known on human somitogenesis as it proceeds between 3- and 5-weeks post conception when embryos are extremely difficult to access. We have developed protocols to differentiate human pluripotent stem cells (ES/iPS) in vitro into PSM. Single cell RNA-sequencing comparison of these human cells differentiating in vitro with mouse embryo PSM reveals that they faithfully recapitulate the PSM differentiation sequence in vitro. Using our in vitro system as a proxy for human somitogenesis\, we were able to demonstrate that human iPS reporter cells harboring a HES7 fluorescent reporter differentiated to PSM exhibit 5-hour oscillations\, thus identifying the human segmentation clock. We have also succeeded in generating PSM organoids that can sequentially form somites exhibiting a normal antero-posterior pattern in vitro. We have taken advantage of these in vitro systems to perform experiments otherwise unfeasible in model organisms. We have generated a human iPS cell line lacking all HOX clusters and demonstrated that while it differentiates to the paraxial mesoderm fate\, it is unable to generate somites. By mimicking key signaling events leading to muscle formation in the embryo\, we developed directed differentiation protocols which recapitulate the developmental sequence of myogenesis. We then used these cells to generate new in vitro models of Duchenne Muscular Dystrophy and to pioneer the production of human satellite cells for cell therapy strategies for muscular dystrophies. Our work provides a framework to study early stages of human myogenesis which are poorly accessible in the embryo.

DESCRIPTION:Invited by the Dumont\, lab Prof. Omaya Dudin (Dpt. Biochemistry\, University of Geneva) will present an Institut Jacques Monod seminar on the theme: \nExploring multicellular developmental programs at the root of animals and beyond \nAbstract: \nAll animals develop from a single-celled zygote and undergo complex morphogenetic processes to form multicellular organisms. These processes are regulated by intrinsic and extrinsic factors that drive key developmental events\, such as symmetry breaking\, cell division\, and differentiation. Despite the remarkable conservation of these pathways across species\, the evolutionary origins of these morphogenetic mechanisms remain unclear. A major challenge in addressing this question is the limited availability of microscopy and cell biological data from key protists that occupy pivotal phylogenetic positions in the eukaryotic tree\, including those identified as the closest unicellular relatives of animals. \nIn this talk\, I will present how our lab is utilizing Expansion Microscopy to capture novel ultrastructural details of microbial life cycles. I will also share recent insights into Ichthyosporeans\, a lineage of protists closely related to animals\, which exhibit diverse multicellular development strategies. These include species that undergo coenocytic development followed by cellularization\, as well as others that develop through cleavage divisions to form spatially organized multicellular colonies. Our findings highlight the importance of studying diverse unicellular taxa to trace the origins of animal multicellularity\, positioning Ichthyosporeans as a promising model for exploring the evolutionary roots of animal embryogenesis.

Programmed DNA elimination (PDE) – a process in which specific cell lineages lose DNA segments or whole chromosomes – represents is a striking deviation to this principle. \n\nPDE is widespread in eukaryotes and plays roles in a variety of cellular processes\, including gene silencing\, germline differentiation\, genome defence\, and non-Mendelian inheritance. It manifests in diverse biological contexts\, including the formation of germline-limited genomes\, meiotic elimination of parental chromosomes\, and sex determination via X-chromosome loss. \nIn recent years\, it has become increasingly clear that PDE occurs across a wide range of phylogenetic groups\, and involves diverse mechanisms.  These findings underscore the overlooked plasticity of genome integrity\, and reveal significant gaps in our understanding of why PDE has evolved repeatedly and is maintained across the Tree of Life. \nThis conference is designed to present the latest research on the mechanisms and evolution of PDE – from the molecular pathways that control genome stability and chromosome segregation\, to genomic conflicts and the long-term evolutionary consequences of PDE on population dynamics and species diversification. We also welcome researchers studying related phenomena\, such as meiotic drive\, B chromosomes\, and those that work on the mechanisms and regulation of genome stability\, chromosome segregation and germline development. \nThe meeting will address the following key topics: \n\n Mechanisms and regulation of genome stability and instability\nMechanisms of chromosome segregation and missegregation\nGenomic conflicts\nEvolutionary dynamics of programmed DNA elimination

DESCRIPTION:Invited by the Konstantinides Lab\, Baptiste Libé Philippot will present an Institut Jacques Monod seminar on the theme: \nCellular and molecular mechanisms underlying human neuronal and circuit evolution \nAbstract: \nBaptiste Libé-Philippot is a CNRS Chargé de recherche and Group Leader at the IBDM (Marseille\, France) since October 2024. He explores the evolution of the development and the properties of human cerebellar neurons and circuits\, with the support of an ERC StG Grant (hCerebEvol). His team “Human Neuronal Evolution” performs cross-species histological comparisons of neuronal morphology\, synaptic innervation and gene expression across the cerebellar development\, in human and other primate species\, to identify human species-specific cellular features. To explore the underlying mechanisms and their functional impact\, his team uses mouse gain-of-function models as well as loss-of-function approaches in human models derived from pluripotent stem cells. This project follows a postdoctoral training in the laboratory of Pierre Vanderhaeghen (VIB – KULeuven\, Belgium) in which he explored the evolutionary divergence of human cortical neurons (Libé-Philippot et al.\, Cell\, 2023 & Neuron\, 2024). This work was honored by the Awards Claude Paoletti (CNRS\, 2024) and Albert Brachet (Royal Academy of Belgium\, 2025).

Artyom Kopp (Department of Evolution and Ecology\, University of California Davis) will present an Institut Jacques Monod on the theme: \nThe Drosophila sex comb as a model of evolutionary innovation: From character identity to cell differentiation \nAbstract: \nThe individualization of serially repeated homologs is one route through which novel traits can evolve. Under this model\, a repeated character—like a limb\, digit\, or sensory bristle—is individualized from its homologs by changes in the regulatory apparatus (‘character identity network’\, ChIN) that specifies its development. Individualization then enables downstream gene networks that build the repeated character to diverge from one another in different parts of the body\, ultimately allowing new phenotypic endpoints to be reached. Despite this model’s intuitive appeal\, the genetic mechanisms through which new ChINs rewire trait-building gene networks remain largely uncharacterized. A promising system in which to study this process is the Drosophila sex comb. Found in a sublineage of Drosophila species\, the sex comb is a recently evolved\, male-specific innovation that evolved from a more evolutionarily ancient precursor—the mechanosensory (MS) bristle—following the gain of a novel ChIN centered on the sex determination gene dsx and HOX gene Scr. We used time-series single-cell RNA-seq to show that rather than co-opting new genes\, this new ChIN orchestrates quantitative and heterochronic changes in the ancestral MS bristle transcriptome. These changes affect gene modules that control endoreplication\, actin dynamics\, and especially energy metabolism. The net effect of these changes is an organ-specific shift in developmental rate\, leading to accelerated growth in sex comb teeth. Our work suggests that morphological innovation can proceed without the co-option of new genes into downstream trait-building networks and instead through metabolically driven differences in developmental rate between serial homologs.

DESCRIPTION:Invited by the Greenberg lab\, Christa Buecker (Associate Professor at Max Perutz Labs\, University Vienna will present an Institut Jacques Monod seminar on the theme: \nBuilding to understand: Enhancer-Promoter Communication in the Context of Regulatory Landscapes \nAbstract: \nEnhancers can regulate promoters from a distance\, but it is still unclear how they establish this  communication. Furthermore\, multiple enhancers often control the activity of the same gene\, yet their interactions remain elusive. We have established a synthetic framework to build regulatory landscapes within the genome of mouse embryonic stem cells to dissect enhancer promoter communication. We have now expanded our toolbox to test enhancer libraries\, alone or in combination with other elements\, to identify principles of distal gene regulation.
