DESCRIPTION:Invited by the Doye Lab\, Leon Peshkin (Mayor-Rotschild visiting professor at the Institut Curie and Principal Research Faculty at the Department of systems biology\, Harvard Medical School) will present an Institut Jacques Monod Seminar on the theme: \n(Phospho)-proteomics of oocyte meiotic division \nAbstract: \nOocyte meiotic divisions represent a critical process in sexual reproduction\, as diploid non-dividing oocyte transforms into a haploid fertilizable egg\, preparing embryonic divisions and differentiation. Although cell differentiation and proliferation are governed by transcription\, oocyte maturation and early embryonic divisions depend entirely on changes in protein abundance and post-translational modifications. Here\, we analyze the abundance and phosphorylation of proteins during Xenopus oocyte meiotic maturation. We reveal significant shifts in protein stability\, related to spindle assembly\, DNA replication and RNA-binding. Our analysis pinpoints broad changes in phosphorylation correlating with key cytological meiotic milestones\, noteworthy changes in membrane trafficking\, nuclear envelope disassembly and modifications in microtubule dynamics. Additionally\, specific phosphorylation events target regulators of protein translation\, Cdk1 and the Mos/MAPK pathway\, providing insights into the dynamics of Cdk1 activity related to the meiotic cell cycle. This study sheds light on the orchestration of protein dynamics and phosphorylation events during oocyte meiotic divisions\, providing a rich resource for understanding the molecular pathways orchestrating meiotic progression in the frog\, and most likely applicable to other vertebrate species. \nBiography: \nLeon Peshkin is a Mayor-Rotschild visiting professor at the Inst Curie and Principal Research Faculty at the Department of systems biology\, Harvard Medical School. His PhD was done in the field of Machine Learning and AI. His eclectic interests range from pharmaco-biology to evo-devo\, to embryology\, to Machine Learning applications in Systems Biology\, but all he does has connection to biology of aging.

SUMMARY:AGRIPOP exhibition - Paleogenetics and archaeology - Neolithic revolution and migration
DESCRIPTION:Exhibition at Université Paris Cité\, Hall A of the Grands-Moulins\, from November 14 to December 16\, 2024 \nThis exhibition opens the doors of the CNRS “Epigenome & Paleogenome” research laboratory at the Institut Jacques Monod\, Université Paris Cité. \nIn 2010\, the first Neanderthal genome was sequenced. This marked the birth of paleogenomics. Since then\, paleogenomics – the analysis of ancient DNA preserved in archaeological bones – has become a rapidly expanding field of research. It provides information on the genetic identity of individuals from the past – humans and animals – on kinship relationships\, genealogies\, migrations\, interbreeding and certain characteristics of their physical appearance. This information enables scientists to reconstruct more accurately the reality of a given moment in the past. \nFrom Çatalhöyük\, in Anatolia\, to the Aisne valley\, the AGRIPOP exhibition invites you to follow the discoveries of archaeologists and paleogeneticists to understand how humans evolved towards the sedentary lifestyle of producers-farmers that is still our present. \nAccessible to all\, this exhibition highlights the collaborative and interdisciplinary practices of researchers on an international scale. Above all\, it makes more tangible our knowledge of the settlement of northern France in the Neolithic by the first farmers of Anatolian origin (modern-day Türkiye) 7\,000 years ago. AGRIPOP sheds new light on the origins of the way our societies function today. The texts in the exhibition are in French but the accompanying booklets are also provided in English.

DESCRIPTION:Invited by the Conduit Lab\, Jens Lüders (Institute for Research in Biomedicine Barcelona) will present an Institut Jacques Monod Seminars on the theme: \nMolecular basis and regulation of microtubule nucleation \n  \nAbstract: \nFormation of new microtubules\, termed nucleation\, is at the heart of organizing\, maintaining\, and remodeling complex microtubule networks. For this reason\, cells tightly control microtubule nucleation in space and time. While microtubules can assemble spontaneously in vitro\, in cells this process requires a nucleator. The main microtubule nucleator is the multi-subunit\, cone-shaped g-tubulin ring complex (gTuRC)\, which has been proposed to nucleate microtubules by a template mechanism. However\, the molecular details underlying its activity and how it is regulated to nucleate microtubules with 13-fold symmetry has remained poorly understood. \nIn my seminar I will present our recent advances in elucidating this process through the generation of recombinant gTuRC and its structural and functional analysis using cryo-EM as well as in vitro and cell-based assays. Key to reconstitution of recombinant gTuRC was the inclusion of the chaperone-like RUVBL1/2 complex in the recombinant expression system\, which greatly improves multi-subunit complex assembly. Surprisingly\, we found that gTuRC has a splayed structure that does not match microtubule symmetry and appears to be a poor nucleation template. However\, when bound to an activator\, the centrosomin motif 1 (CM1) of the gTuRC adapter protein CDK5RAP2\, gTuRC acquires the ability to adopt microtubule-like conformations and becomes an efficient nucleator. Activation involves binding of multiple CM1 dimers to five distinct sites around the outside of the gTuRC cone\, which modulates lateral interactions among the GCP subunits that form the cone-like structure. This facilitates microtubule-like conformations and release of luminal actin that is integral to non-activated gTuRC. \nWe propose a model where generation of gTuRC with an expanded conformational range\, rather than perfect symmetry\, is sufficient to boost nucleation activity.
