SUMMARY:Institut Jacques Monod lecture - Jordan Raff
DESCRIPTION:Invited by the Institut Jacques Monod\, Prof Jordan Raff\, (Sir William Dunn School of Pathology\, University of Oxford) will present an Institut Jacques Monod lecture: \nHow do cells build complicated organelles? \nAbstract: \nA key feature of all living things is their ability to assemble complicated structures using dilute building blocks present in the cell. How cells achieve this remarkable feat of bioengineering is largely unknown. Centrosomes are major microtubule (MT) organising centres and important signalling hubs in many cells. They are an excellent model for studying organelle biogenesis as they comprise 10s-100s of copies of several hundred different types of protein yet\, like the DNA\, they precisely duplicate once per cell cycle. We have recently reconstituted centrosome duplication on synthetic beads programmed to recruit core centrosome assembly proteins when injected into Drosophila embryos. These beads generate structures that appear to be functionally indistinguishable from centrosomes: they recruit centriole/centrosome proteins\, organise MTs and proceed through multiple rounds of high-fidelity duplication\, all in synchrony with the endogenous centrosomes. The beads function as seeds that recruit biogenesis-promoting proteins to stimulate the assembly of relatively simple scaffolds that direct centrosome self-assembly. This “Seed—Scaffold—Self-Assemble” mechanism may represent a general principle of organelle biogenesis\, explaining how simple molecular inputs can generate complex structures without the need to copy a pre-existing template.

DESCRIPTION:Invited by the Ladoux/Mege lab\, Bram Hoogland (Vrije Universiteit\, Amsterdam\, Netherlands) will present an Institut Jacques Monod seminar on the theme: \nData-Driven Inference of Cell Migration and Polarity Dynamics \nAbstract: \nCell migration emerges from coupled processes including polarity\, biochemical signaling\, adhesion\, and mechanical interactions. These processes govern how cells move\, deform\, and respond to one another\, yet integrating them into a unified dynamical description remains a major challenge. Experiments typically capture only the resulting behaviors\, such as cell shape and trajectories. Here\, we present a data-driven framework to learn and disentangle underlying dynamics directly from observed cell behavior and apply it across scales from polarization dynamics of single cells to multicellular systems. In multicellular migration\, our framework reveals distinct interaction rules across cell types. Non-cancerous epithelial cells exhibit reciprocal repulsion and velocity alignment\, whereas cancerous mesenchymal cells additionally show nonreciprocal interactions that modulate self-propulsion. These nonreciprocal interactions shift the onset and speed of cellular flocks\, linking microscopic interaction rules to collective behavior. Using experiments with a molecular polarity marker\, (from the amazing Joseph)\, we uncover how cell motion\, morphology\, and polarity are coupled. We identify dynamical states associated with persistent migration in small cells but find that these states are absent in large cells\, revealing a coupling between cell length and polarity-driven migration. Lastly\, we have started making the first steps in learning a full 3D stochastic reaction diffusion model directly from experiments on budding yeast\, capturing their symmetry breaking and finding active transport. Overall\, our data-driven framework provides a principled route to learning interpretable dynamics from experiments\, enabling comparison across cell types and linking cell-intrinsic dynamics and cell-cell interactions to emergent behavior.

DESCRIPTION:Invited by the Conduit lab\, Claudia Brito (Center for Genomic Regulation (CRG\, Barcelona)\, incoming CNRS ATIP Group Leader\, Institute for Integrative Biology of the Cell (I2BC)\, Paris-Saclay) will present an Institut Jacques Monod serminar on the theme: \nMechanisms regulating γTuRC-mediated microtubule nucleation \nAbstract: \nThe γ-tubulin ring complex (γTuRC) is the principal microtubule nucleator in cells and must therefore be tightly regulated to ensure that nucleation occurs at the right time and place. \nRecently\, we found that this regulation relies on a structural transition of γTuRC from an open to a closed state\, where the complex matches microtubule geometry. In cells\, molecules containing a specific centrosomal domain called Centrosomin Motif 1 (CM1) are known to activate the complex\, but the molecular mechanism underlying this activation has remained unknown. \nIn this presentation\, I will discuss how we combined in vitro reconstitution using TIRF microscopy with cryo-EM to dissect the mechanisms of γTuRC closure and CM1-mediated activation\, providing insight into this important process in cells.

DESCRIPTION:Invited by the Institut Jacques Monod\, Silvia Torchio (Fate and plasticity of epithelial stem cells Lab\, Developmental Biology and Genetics Unit (UMR3215/U934)\, Institut Curie) will present a Paris Postdoc Seminar on the theme: \nThe gut at a cROSsroad: how Reactive Oxygen Species shape cell fate decisions \nAbstract: \nStem cells have to continuously balance between self-renewal and differentiation. Although we know a great deal about the genetic\, epigenetic\, and mechanical cues controlling stem cell fate decisions\, the contribution of biochemical signaling such as Reactive Oxygen Species (ROS)-mediated is still poorly understood. We thus investigated redox as a potential mechanism governing stem cell fate decisions in the context of the murine small intestine\, leveraging organoids as a model. We determined that ROS concentration is higher in stem cells than in differentiated lineages\, both in vitro and ex vivo; this difference is crucial to maintain balanced cell type composition in organoids\, as ROS alterations skew stem cell differentiation versus self-renewal equilibrium. Moreover\, increasing ROS levels in differentiated Paneth cells triggers their reentry into the cell cycle\, in a process akin to regeneration. Furthermore\, we show that a ROS spike is crucial during development for the maturation of fetal progenitors into the adult stem cell pool. Collectively\, our work shows that redox signaling acts as a conserved mechanism to regulate stem cell fate decisions in the mouse small intestine\, both during development and in adult homeostasis.

DESCRIPTION:Invited by the Dumont\, lab Prof. Omaya Dudin (Dpt. Biochemistry\, University of Geneva) will present an Institut Jacques Monod seminar on the theme: \nExploring multicellular developmental programs at the root of animals and beyond \nAbstract: \nAll animals develop from a single-celled zygote and undergo complex morphogenetic processes to form multicellular organisms. These processes are regulated by intrinsic and extrinsic factors that drive key developmental events\, such as symmetry breaking\, cell division\, and differentiation. Despite the remarkable conservation of these pathways across species\, the evolutionary origins of these morphogenetic mechanisms remain unclear. A major challenge in addressing this question is the limited availability of microscopy and cell biological data from key protists that occupy pivotal phylogenetic positions in the eukaryotic tree\, including those identified as the closest unicellular relatives of animals. \nIn this talk\, I will present how our lab is utilizing Expansion Microscopy to capture novel ultrastructural details of microbial life cycles. I will also share recent insights into Ichthyosporeans\, a lineage of protists closely related to animals\, which exhibit diverse multicellular development strategies. These include species that undergo coenocytic development followed by cellularization\, as well as others that develop through cleavage divisions to form spatially organized multicellular colonies. Our findings highlight the importance of studying diverse unicellular taxa to trace the origins of animal multicellularity\, positioning Ichthyosporeans as a promising model for exploring the evolutionary roots of animal embryogenesis.

Programmed DNA elimination (PDE) – a process in which specific cell lineages lose DNA segments or whole chromosomes – represents is a striking deviation to this principle. \n\nPDE is widespread in eukaryotes and plays roles in a variety of cellular processes\, including gene silencing\, germline differentiation\, genome defence\, and non-Mendelian inheritance. It manifests in diverse biological contexts\, including the formation of germline-limited genomes\, meiotic elimination of parental chromosomes\, and sex determination via X-chromosome loss. \nIn recent years\, it has become increasingly clear that PDE occurs across a wide range of phylogenetic groups\, and involves diverse mechanisms.  These findings underscore the overlooked plasticity of genome integrity\, and reveal significant gaps in our understanding of why PDE has evolved repeatedly and is maintained across the Tree of Life. \nThis conference is designed to present the latest research on the mechanisms and evolution of PDE – from the molecular pathways that control genome stability and chromosome segregation\, to genomic conflicts and the long-term evolutionary consequences of PDE on population dynamics and species diversification. We also welcome researchers studying related phenomena\, such as meiotic drive\, B chromosomes\, and those that work on the mechanisms and regulation of genome stability\, chromosome segregation and germline development. \nThe meeting will address the following key topics: \n\n Mechanisms and regulation of genome stability and instability\nMechanisms of chromosome segregation and missegregation\nGenomic conflicts\nEvolutionary dynamics of programmed DNA elimination

DESCRIPTION:Invited by the Konstantinides Lab\, Baptiste Libé Philippot will present an Institut Jacques Monod seminar on the theme: \nCellular and molecular mechanisms underlying human neuronal and circuit evolution \nAbstract: \nBaptiste Libé-Philippot is a CNRS Chargé de recherche and Group Leader at the IBDM (Marseille\, France) since October 2024. He explores the evolution of the development and the properties of human cerebellar neurons and circuits\, with the support of an ERC StG Grant (hCerebEvol). His team “Human Neuronal Evolution” performs cross-species histological comparisons of neuronal morphology\, synaptic innervation and gene expression across the cerebellar development\, in human and other primate species\, to identify human species-specific cellular features. To explore the underlying mechanisms and their functional impact\, his team uses mouse gain-of-function models as well as loss-of-function approaches in human models derived from pluripotent stem cells. This project follows a postdoctoral training in the laboratory of Pierre Vanderhaeghen (VIB – KULeuven\, Belgium) in which he explored the evolutionary divergence of human cortical neurons (Libé-Philippot et al.\, Cell\, 2023 & Neuron\, 2024). This work was honored by the Awards Claude Paoletti (CNRS\, 2024) and Albert Brachet (Royal Academy of Belgium\, 2025).

SUMMARY:Institut Jacques Monod seminar - Christa Buecker
DESCRIPTION:Invited by the Greenberg lab\, Christa Buecker (Associate Professor at Max Perutz Labs\, University Vienna will present an Institut Jacques Monod seminar on the theme: \nBuilding to understand: Enhancer-Promoter Communication in the Context of Regulatory Landscapes \nAbstract: \nEnhancers can regulate promoters from a distance\, but it is still unclear how they establish this  communication. Furthermore\, multiple enhancers often control the activity of the same gene\, yet their interactions remain elusive. We have established a synthetic framework to build regulatory landscapes within the genome of mouse embryonic stem cells to dissect enhancer promoter communication. We have now expanded our toolbox to test enhancer libraries\, alone or in combination with other elements\, to identify principles of distal gene regulation.

SUMMARY:Ctrl+Epi+Edit\, Engineering the Epigenome
DESCRIPTION:Epigenome editing is a rapidly emerging field that has the potential to revolutionise genomic medicine by offering new ways to precisely program gene expression and treat a range of diseases. Precision control of the epigenome and gene activity is further unlocking key mechanistic insights into a wide variety of molecular and cellular processes. The potential of epigenome editing technologies for both research and therapeutic applications has thus led to widespread excitement over the last five years\, in areas ranging from neurobiology\, to high-throughput (epi)genomics to agriculture. As the technologies continue to emerge and the epigenome editing field begins to coalesce\, we feel now is the time to bring interdisciplinary experts and leaders in the field together. This will enable\, for the first time\, epigenome editors and related disciplines to collectively discuss the technology\, its applications and ethics\, and the exciting scientific insights being generated. Such a symposium will help to accelerate progress in this field by facilitating collaborations between researchers and providing a platform for the dissemination of new discoveries and techniques.
