How cells achieve this remarkable feat of bioengineering is largely unknown. Centrosomes are major microtubule (MT) organising centres and important signalling hubs in many cells. They are an excellent model for studying organelle biogenesis as they comprise 10s-100s of copies of several hundred different types of protein yet\, like the DNA\, they precisely duplicate once per cell cycle. We have recently reconstituted centrosome duplication on synthetic beads programmed to recruit core centrosome assembly proteins when injected into Drosophila embryos. These beads generate structures that appear to be functionally indistinguishable from centrosomes: they recruit centriole/centrosome proteins\, organise MTs and proceed through multiple rounds of high-fidelity duplication\, all in synchrony with the endogenous centrosomes. The beads function as seeds that recruit biogenesis-promoting proteins to stimulate the assembly of relatively simple scaffolds that direct centrosome self-assembly. This “Seed—Scaffold—Self-Assemble” mechanism may represent a general principle of organelle biogenesis\, explaining how simple molecular inputs can generate complex structures without the need to copy a pre-existing template. These processes govern how cells move\, deform\, and respond to one another\, yet integrating them into a unified dynamical description remains a major challenge. Experiments typically capture only the resulting behaviors\, such as cell shape and trajectories. Here\, we present a data-driven framework to learn and disentangle underlying dynamics directly from observed cell behavior and apply it across scales from polarization dynamics of single cells to multicellular systems. In multicellular migration\, our framework reveals distinct interaction rules across cell types. Non-cancerous epithelial cells exhibit reciprocal repulsion and velocity alignment\, whereas cancerous mesenchymal cells additionally show nonreciprocal interactions that modulate self-propulsion. These nonreciprocal interactions shift the onset and speed of cellular flocks\, linking microscopic interaction rules to collective behavior. Using experiments with a molecular polarity marker\, (from the amazing Joseph)\, we uncover how cell motion\, morphology\, and polarity are coupled. We identify dynamical states associated with persistent migration in small cells but find that these states are absent in large cells\, revealing a coupling between cell length and polarity-driven migration. Lastly\, we have started making the first steps in learning a full 3D stochastic reaction diffusion model directly from experiments on budding yeast\, capturing their symmetry breaking and finding active transport. Overall\, our data-driven framework provides a principled route to learning interpretable dynamics from experiments\, enabling comparison across cell types and linking cell-intrinsic dynamics and cell-cell interactions to emergent behavior. In cells\, molecules containing a specific centrosomal domain called Centrosomin Motif 1 (CM1) are known to activate the complex\, but the molecular mechanism underlying this activation has remained unknown. \nIn this presentation\, I will discuss how we combined in vitro reconstitution using TIRF microscopy with cryo-EM to dissect the mechanisms of γTuRC closure and CM1-mediated activation\, providing insight into this important process in cells. Although we know a great deal about the genetic\, epigenetic\, and mechanical cues controlling stem cell fate decisions\, the contribution of biochemical signaling such as Reactive Oxygen Species (ROS)-mediated is still poorly understood. We thus investigated redox as a potential mechanism governing stem cell fate decisions in the context of the murine small intestine\, leveraging organoids as a model. We determined that ROS concentration is higher in stem cells than in differentiated lineages\, both in vitro and ex vivo; this difference is crucial to maintain balanced cell type composition in organoids\, as ROS alterations skew stem cell differentiation versus self-renewal equilibrium. Moreover\, increasing ROS levels in differentiated Paneth cells triggers their reentry into the cell cycle\, in a process akin to regeneration. Furthermore\, we show that a ROS spike is crucial during development for the maturation of fetal progenitors into the adult stem cell pool. Collectively\, our work shows that redox signaling acts as a conserved mechanism to regulate stem cell fate decisions in the mouse small intestine\, both during development and in adult homeostasis. These structures form periodically from a posterior tissue called Presomitic Mesoderm (PSM). The rhythmic formation of somites involves a molecular oscillator called segmentation clock which drives pulses of Notch\, Wnt and FGF signaling in the PSM. Virtually nothing is known on human somitogenesis as it proceeds between 3- and 5-weeks post conception when embryos are extremely difficult to access. We have developed protocols to differentiate human pluripotent stem cells (ES/iPS) in vitro into PSM. Single cell RNA-sequencing comparison of these human cells differentiating in vitro with mouse embryo PSM reveals that they faithfully recapitulate the PSM differentiation sequence in vitro. Using our in vitro system as a proxy for human somitogenesis\, we were able to demonstrate that human iPS reporter cells harboring a HES7 fluorescent reporter differentiated to PSM exhibit 5-hour oscillations\, thus identifying the human segmentation clock. We have also succeeded in generating PSM organoids that can sequentially form somites exhibiting a normal antero-posterior pattern in vitro. We have taken advantage of these in vitro systems to perform experiments otherwise unfeasible in model organisms. We have generated a human iPS cell line lacking all HOX clusters and demonstrated that while it differentiates to the paraxial mesoderm fate\, it is unable to generate somites. By mimicking key signaling events leading to muscle formation in the embryo\, we developed directed differentiation protocols which recapitulate the developmental sequence of myogenesis. We then used these cells to generate new in vitro models of Duchenne Muscular Dystrophy and to pioneer the production of human satellite cells for cell therapy strategies for muscular dystrophies. Our work provides a framework to study early stages of human myogenesis which are poorly accessible in the embryo. Biochemistry\, University of Geneva) will present an Institut Jacques Monod seminar on the theme: \nExploring multicellular developmental programs at the root of animals and beyond \nAbstract: \nAll animals develop from a single-celled zygote and undergo complex morphogenetic processes to form multicellular organisms. These processes are regulated by intrinsic and extrinsic factors that drive key developmental events\, such as symmetry breaking\, cell division\, and differentiation. Despite the remarkable conservation of these pathways across species\, the evolutionary origins of these morphogenetic mechanisms remain unclear. A major challenge in addressing this question is the limited availability of microscopy and cell biological data from key protists that occupy pivotal phylogenetic positions in the eukaryotic tree\, including those identified as the closest unicellular relatives of animals. \nIn this talk\, I will present how our lab is utilizing Expansion Microscopy to capture novel ultrastructural details of microbial life cycles. I will also share recent insights into Ichthyosporeans\, a lineage of protists closely related to animals\, which exhibit diverse multicellular development strategies. These include species that undergo coenocytic development followed by cellularization\, as well as others that develop through cleavage divisions to form spatially organized multicellular colonies. Our findings highlight the importance of studying diverse unicellular taxa to trace the origins of animal multicellularity\, positioning Ichthyosporeans as a promising model for exploring the evolutionary roots of animal embryogenesis. Programmed DNA elimination (PDE) – a process in which specific cell lineages lose DNA segments or whole chromosomes – represents is a striking deviation to this principle. \n\nPDE is widespread in eukaryotes and plays roles in a variety of cellular processes\, including gene silencing\, germline differentiation\, genome defence\, and non-Mendelian inheritance. It manifests in diverse biological contexts\, including the formation of germline-limited genomes\, meiotic elimination of parental chromosomes\, and sex determination via X-chromosome loss. \nIn recent years\, it has become increasingly clear that PDE occurs across a wide range of phylogenetic groups\, and involves diverse mechanisms.  These findings underscore the overlooked plasticity of genome integrity\, and reveal significant gaps in our understanding of why PDE has evolved repeatedly and is maintained across the Tree of Life. \nThis conference is designed to present the latest research on the mechanisms and evolution of PDE – from the molecular pathways that control genome stability and chromosome segregation\, to genomic conflicts and the long-term evolutionary consequences of PDE on population dynamics and species diversification. We also welcome researchers studying related phenomena\, such as meiotic drive\, B chromosomes\, and those that work on the mechanisms and regulation of genome stability\, chromosome segregation and germline development. \nThe meeting will address the following key topics: \n\n Mechanisms and regulation of genome stability and instability\nMechanisms of chromosome segregation and missegregation\nGenomic conflicts\nEvolutionary dynamics of programmed DNA elimination URL:https://www.ijm.fr/event/conference-jacques-monod-the-mechanistic-and-evolutionary-basis-of-programmed-dna-elimination/?lang=en LOCATION:Roscoff\, Roscoff\, France ATTACH;FMTTYPE=image/jpeg:https://www.ijm.fr/wp-content/uploads/2026/03/SD-1-scaled.jpg END:VEVENT BEGIN:VEVENT DTSTART;TZID=Europe/Paris:20260921T140000 DTEND;TZID=Europe/Paris:20260921T170000 DTSTAMP:20260817T131742Z CREATED:20260821T080055Z LAST-MODIFIED:20260817T131742Z UID:33726-1789999200-1790010000@www.ijm.fr SUMMARY:PhD thesis defense - Rebekah Ricquebourg DESCRIPTION:Rebekah Ricquebourg (Konstantinides Lab ) will defend her PhD thesis: \nSpatial factors affect temporal patterning of neuronal stem cells to regulate neuronal diversity \nThe jury will be composed of: \n\nAriane RAMAEKERS : Rapporteuse\nJonathan ENRIQUEZ : Rapporteur\nPauline SPÉDER : Examinatrice\nMuriel PERRON : Examinatrice\nBaptiste LIBÉ-PHILIPPOT : Examinateur\nNikolaos KONSTANTINIDES : Directeur de thèse URL:https://www.ijm.fr/event/phd-thesis-defense-rebekah-ricquebourg/?lang=en LOCATION:Bâtiment Condorcet Amphithéâtre Pierre Gilles de Gennes\, 4 rue Elsa Morante\, Paris\, Sélectionner un État :\, 75013\, France ATTACH;FMTTYPE=image/jpeg:https://www.ijm.fr/wp-content/uploads/2026/07/These-bandeau-Rebekah-Ricquebourg-scaled.jpg END:VEVENT BEGIN:VEVENT DTSTART;TZID=Europe/Paris:20260922T114500 DTEND;TZID=Europe/Paris:20260922T130000 DTSTAMP:20260814T083439Z CREATED:20260814T083439Z LAST-MODIFIED:20260814T083439Z UID:33909-1790077500-1790082000@www.ijm.fr SUMMARY:Institut Jacques Monod seminar- Baptiste Libé Philippot DESCRIPTION:Invited by the Konstantinides Lab\, Baptiste Libé Philippot will present an Institut Jacques Monod seminar on the theme: \nCellular and molecular mechanisms underlying human neuronal and circuit evolution \nAbstract: \nBaptiste Libé-Philippot is a CNRS Chargé de recherche and Group Leader at the IBDM (Marseille\, France) since October 2024. Artyom Kopp (Department of Evolution and Ecology\, University of California Davis) will present an Institut Jacques Monod on the theme: \nThe Drosophila sex comb as a model of evolutionary innovation: From character identity to cell differentiation \nAbstract: \nThe individualization of serially repeated homologs is one route through which novel traits can evolve. Under this model\, a repeated character—like a limb\, digit\, or sensory bristle—is individualized from its homologs by changes in the regulatory apparatus (‘character identity network’\, ChIN) that specifies its development. Individualization then enables downstream gene networks that build the repeated character to diverge from one another in different parts of the body\, ultimately allowing new phenotypic endpoints to be reached. Despite this model’s intuitive appeal\, the genetic mechanisms through which new ChINs rewire trait-building gene networks remain largely uncharacterized. A promising system in which to study this process is the Drosophila sex comb. Found in a sublineage of Drosophila species\, the sex comb is a recently evolved\, male-specific innovation that evolved from a more evolutionarily ancient precursor—the mechanosensory (MS) bristle—following the gain of a novel ChIN centered on the sex determination gene dsx and HOX gene Scr. We used time-series single-cell RNA-seq to show that rather than co-opting new genes\, this new ChIN orchestrates quantitative and heterochronic changes in the ancestral MS bristle transcriptome. These changes affect gene modules that control endoreplication\, actin dynamics\, and especially energy metabolism. The net effect of these changes is an organ-specific shift in developmental rate\, leading to accelerated growth in sex comb teeth. Our work suggests that morphological innovation can proceed without the co-option of new genes into downstream trait-building networks and instead through metabolically driven differences in developmental rate between serial homologs. For instance\, the extraembryonic amnion\, which forms a fluid-filled sac surrounding the embryo\, was recently shown to trigger germ layer specification during gastrulation\, by secreting BMP ligands. Despite the key roles of extraembryonic tissues in embryo development\, little is still known regarding their molecular and biophysical programs\, particularly in human. Using a 2D stem cell-based model of human gastrulation\, termed gastruloid discs\, we found that amnion cells undergo a sharp columnar-to-squamous transition concomitantly with fate specification. Via biophysical modelling\, direct force measurements\, pharmacological and genetic perturbations\, we showed that this morphogenetic transition is amnion-intrinsic and it is driven by active wetting\, i.e. a transition from tensile to adhesion-dominated cellular states. Molecularly\, active wetting is implemented via a rewiring of cytoskeleton composition\, from actomyosin to keratin-based cytoskeletal networks\, akin to a bistable toggle-switch in gene regulatory networks. Strikingly\, blocking shape changes at the colony edge results both in defective cellular states in the amnion and impaired gastruloid disc morphogenesis within the embryonic compartment. Together\, our findings establish that a cytoskeletal toggle switch couples fate specification to tissue architecture in the human amnion and suggest an unexpectedly active mechanical role for extraembryonic tissues in shaping the embryo proper. Those cells also constantly proliferate and differentiate into dozens of different cell types to maintain tissue homeostasis. However\, how cell fate acquisition is coordinated with cell cycle progression remains poorly understood. We choose to address this question in Schmidtea mediterranea using single cell transcriptomics. We generated scRNA-seq datasets from cells enriched for 2C or 4C DNA content and from animals subjected to RNAi against the cell cycle regulators cdh1 and h2b. Our analyses show that progenitor populations across multiple lineages contain actively cycling cells and that lineage commitment occurs at different stages depending on the cell type. Comparative analyses reveal that Schmidtea possesses a simplified repertoire of canonical cell cycle regulators\, consistent with a central role for cdh1 in controlling cell cycle exit. Functional perturbation of cdh1 demonstrates that cell cycle exit is needed for differentiation but not for lineage specification. Together\, these findings provide new insights into how cell cycle regulation is coordinated with lineage commitment and differentiation in planarian stem cells. Jens Januschke DESCRIPTION:Invited by the Guichet lab\, Jens Januschke ( Faculty of Life at the University of Dundee\, Scotland\, UK) will present an Institut Jacques Monod on the theme: \nWhen size determines fate: rethinking asymmetric stem cell division of neural stem cells of the fly \nAbstract: \nHow does a stem cell produce two daughters with different identities? Classical models emphasise that in intrinsically controlled asymmetric cell division\, unequal inheritance of molecules can determine fate\, ensuring for instance in some systems that one daughter retains stem cell identity while the other embarks on differentiation. But what if something as simple as cell size could override these molecular instructions? \nAsymmetric cell division is a fundamental mechanism of development and tissue homeostasis. In Drosophila neural stem cells\, this process generates daughters that differ in fate but also dramatically in size. Understanding how this physical asymmetry emerges reveals an intricate interplay between cell polarity\, cortical mechanics and membrane dynamics. Spatially controlled actomyosin contractility and local membrane expansion work together to shape dividing cells\, translating molecular organisation into differences in daughter-cell size. \nIntriguingly\, cell size is more than a passive consequence of asymmetric division. It can actively influence developmental decisions\, even challenging the instructions provided by inherited fate determinants. This raises the possibility that a cell’s physical characteristics are integral to how its identity is established and maintained. \nIn this seminar I will summarise our findings and sketch our direction of travel on the regulation of cell size asymmetry and its contribution on cell fate. \n  \nJens Januschke is joint Head of Molecular\, Cell & Developmental Biology in the Faculty of Life at the University of Dundee. He studied at the University of Cologne and obtained his PhD in genetics from the University of Paris VI before joining the Institute for Research in Biomedicine in Barcelona as a postdoc. Jens started his independent research at the University of Dundee for which he was awarded a Sir Henry Dale fellowship in 2013. In 2023 Jens was awarded a personal chair in Developmental Cell Biology at the University of Dundee. \nHis group is interested in fundamental aspects of cell function during development. He uses Drosophila as a model system with a focus on epithelial biology and epithelial derived asymmetrically dividing neural stem cells of the developing central nervous system. His group uses genetics\, chemical genetics and live cell imaging to unravel principles of how the cell polarity machinery controls epithelial mechanics as well as cell fate decisions in the developing nervous system. Within this context\, our lab is interested in a type of epigenetic marks named histone acylations\, whose abundance depends upon the availability of their precursor metabolites and\, hence\, are sensitive to the cell metabolic status. Among the different histone acylations\, we have recently characterized that lactylation modifies several histone lysine residues in wild-type mouse testis by proteomics approaches (Manessier et al.\, 2026; PMID 42285509). \nTo better understand the effects of a metabolic stress on coordinated metabolo-epigenetic signatures in male germ cells\, we established a mouse model of Western style diet-induced obesity (WD; high-fat/high-sugar\, 16 weeks). WD males exhibit an altered testicular metabolome\, characterized by changes in pathways related to energy metabolism and the generation of precursors of epigenetic marks. In parallel\, we identified altered histone acylation levels in male germ cells of WD mice\, suggesting a link between the observed metabolomic and epigenetic alterations. Multi-omics approaches in WD mice are currently being analyzed to determine the sensitivity to metabolic changes in male germ cells. \nThus\, our work supports that the testis can sense metabolic disturbances\, leading to dysregulations in developing male germ cells at the metabolic and epigenetic levels. These alterations could contribute to the effects of male obesity beyond fertilization described in the POHaD framework.
