SUMMARY:Paris Postdoc Seminar - Alberto De La Iglesia
DESCRIPTION:Invited by the Institut Jacques Monod\, Alberto de la Iglesia\, Ph.D. Institut Cochin (Inserm U1016 – CNRS UMR8104 – Université Paris Cité) Dr. Julie Cocquet Group “Epigenetics & Spermatogenesis”\, part of the VAIMAN Team “From Gametes to Birth: Genomics\, Epigenetics\, and Physiopathology of Reproduction” will present a Paris Postdoc Seminar on te theme : \nExploring the role of metabolism-dependent histone acylations during male germ cell differentiation and their sensitivity to metabolic perturbations  \nAbstract: \nThe paternal epigenome is established during spermatogenesis and is particularly responsive to environmental cues. In line with the POHaD paradigm (Paternal Origins of Health and Disease)\, increasing evidence shows that environmentally induced paternal epigenetic alterations can have effects beyond fertilization\, which is the case of paternal obesity. Within this context\, our lab is interested in a type of epigenetic marks named histone acylations\, whose abundance depends upon the availability of their precursor metabolites and\, hence\, are sensitive to the cell metabolic status. Among the different histone acylations\, we have recently characterized that lactylation modifies several histone lysine residues in wild-type mouse testis by proteomics approaches (Manessier et al.\, 2026; PMID 42285509). \nTo better understand the effects of a metabolic stress on coordinated metabolo-epigenetic signatures in male germ cells\, we established a mouse model of Western style diet-induced obesity (WD; high-fat/high-sugar\, 16 weeks). WD males exhibit an altered testicular metabolome\, characterized by changes in pathways related to energy metabolism and the generation of precursors of epigenetic marks. In parallel\, we identified altered histone acylation levels in male germ cells of WD mice\, suggesting a link between the observed metabolomic and epigenetic alterations. Multi-omics approaches in WD mice are currently being analyzed to determine the sensitivity to metabolic changes in male germ cells. \nThus\, our work supports that the testis can sense metabolic disturbances\, leading to dysregulations in developing male germ cells at the metabolic and epigenetic levels. These alterations could contribute to the effects of male obesity beyond fertilization described in the POHaD framework.

SUMMARY:Institut Jacques Monod seminar - Philipp Brand
DESCRIPTION:Invited by the Konstantinides lab\, Philipp Brand (Group leader of the Evolutionary Neuroscience and Behavior Group\, Max Planck Institute for Brain Research): \nBehavioral evolution in a complex world: From environment to neural circuits \nAbstract: \nFrom dancing birds to singing flies\, animals have evolved an astounding diversity of behaviors to attract and choose mating partners. Guided by innate preferences and aversions\, the brain filters complex environments for cues and signals to successfully navigate these reproductive behaviors in appropriate spatial\, temporal\, and social contexts. To understand how the natural environment shapes sensory evolution and contributes to the diversification of neural circuits controlling mating behaviors\, my lab studies the highly tractable nervous system of Drosophila as an inroad. \nIn Drosophila\, courtship and mating occurs on fermenting food where many individuals congregate. Replicating this naturalistic context in the lab\, we discovered striking differences in the sensory dependence of mating behaviors across species. Most notably\, the host specialist Drosophila erecta requires the presence of food to engage in courtship and only mates in social groups. Harnessing the power of cross-species neurogenetics\, we reveal that sexual arousal in D. erecta is uniquely gated by food odors that switch the valence of visual stimuli driving courtship. This provides a possible neural mechanism underlying the environmental modulation of reproductive behaviors\, highlighting how social behaviors are shaped by the natural environments in which they evolve.

SUMMARY:Ctrl+Epi+Edit\, Engineering the Epigenome
DESCRIPTION:Epigenome editing is a rapidly emerging field that has the potential to revolutionise genomic medicine by offering new ways to precisely program gene expression and treat a range of diseases. Precision control of the epigenome and gene activity is further unlocking key mechanistic insights into a wide variety of molecular and cellular processes. The potential of epigenome editing technologies for both research and therapeutic applications has thus led to widespread excitement over the last five years\, in areas ranging from neurobiology\, to high-throughput (epi)genomics to agriculture. As the technologies continue to emerge and the epigenome editing field begins to coalesce\, we feel now is the time to bring interdisciplinary experts and leaders in the field together. This will enable\, for the first time\, epigenome editors and related disciplines to collectively discuss the technology\, its applications and ethics\, and the exciting scientific insights being generated. Such a symposium will help to accelerate progress in this field by facilitating collaborations between researchers and providing a platform for the dissemination of new discoveries and techniques. It will also help to raise broader awareness of this exciting emerging science and its potential to transform precision medicine. \n\n\n\n\nTime and Place\n\nThe meeting will take place from November 16-17  at Institut Jacques Monod in the heart of Paris\, France \nWe will have two half day sessions\, and a poster session over cocktails and light bites \n\n\n\nKey Dates\n\n\nAbstract Submission: October 1\, 2026 \nRegistration: November 9\, 2026 \n\n\n\n\nInvited Speakers\n\n🧬 Gabriella Ficz (QMUL\, UK) \n🧬 Charles Gersbach (Duke University\, USA) \n🧬 Jamie Hackett (EMBL Rome\, Italy) \n🧬 Jake Harris (Cambridge University\, UK) \n🧬 Angelo Lombardo (San Raffaele University\, Italy) \n🧬 Reini Luco (Institut Curie\, France) \n🧬 Mariane Rots (UMCG\, Netherlands) \n🧬 Edda Schulz (MPI Berlin\, Germany) \n🧬 Stefan Stricker (Helmholtz Munich\, Germany) \nMore speakers will be selected from abstracts! \nYou can find all the informations regarding the symposium\, the registration and abstract submission here  \nhttp://www.ijm.fr/wp-content/uploads/2026/03/affiche.jpg

SUMMARY:Institut Jacques Monod Seminar - Prof. Karin Forsberg-Nilsson
DESCRIPTION:Invited by the Ladoux/Mège\, Prof. Karin Forsberg-Nilsson (Uppsala University\, Sweden) will present an Institut Jacques Monod on the theme: \nLeveraging non-coding mutations with evolutionary constraint to discover novel driver genes in solid tumors \nAbstract: \nCancer genomics has been biased against non-coding mutations\, which account for 98% of all mutations\, and driver mutations outside exons remain underrepresented. We hypothesized that evolutionary constraint could be used to identify functional cancer mutations. In our previous work\, we successfully identified functionally active non-coding constraint mutations (NCCMs) in glioblastoma. The Zoonomia Consortium has since published single-base (phyloP) constraint scores from whole-genome alignments of 240 mammals. Through comparisons with previous large-scale genome annotations and datasets such as ClinVar\, we provide proof of concept that evolutionarily constrained positions are enriched for disease-associated variants\, including those implicated in medulloblastoma. \nWe next examined whether NCCMs and their associated genes exert functional effects across multiple tumor types. To investigate this\, we mapped evolutionary constraint annotations to somatic single-nucleotide variants (SNVs) and insertions/deletions across more than 2\,500 tumors representing 19 tumor types in the ICGC-PCAWG dataset. This analysis identified 56 pan-cancer and 132 cancer-type-specific putative driver genes enriched for NCCMs. Notably\, genes with a high NCCM load rarely harbor protein-coding mutations\, suggesting that distinct selective pressures preserve protein integrity while altering gene expression regulation. \nCollectively\, these findings demonstrate the power of evolutionary constraint to decode non-coding genome function and identify candidate regulatory drivers of cancer. \nBiography : \nKarin Forsberg-Nilsson is Professor of Stem Cell Research in the Department of Immunology\, Genetics and Pathology at Uppsala University\, Sweden. Her research brings together stem cell biology and cancer genomics\, with a focus on brain tumors and the identification of cancer driver genes. She obtained her PhD at Uppsala University in 1992 and conducted postdoctoral research at the US National Institutes of Health. She served as Head of Department from 2014 to 2020 and Dean of the Faculty of Medicine from 2020 to 2026. In 2015\, she was a visiting professor at the Collège de France.

SUMMARY:Institut Jacques Monod seminar - Idoia Quintana Urzainqui
DESCRIPTION:Invitéed by the Konstantinides lab\, Idoia Quintana Urzainqui ( Université Paris Saclay\, France) will present an Institut Jacques Monod seminar on the theme: \nStudying the shark embryo at the single cell level to understand the origin and evolution of the vertebrate telencephalon \nAbstract: \nMuch of what we know about vertebrate brain evolution comes from studying mammals. Cartilaginous fish\, such as sharks\, have been largely missing from this picture. They emerged around 450 million years ago\, at a pivotal transition in vertebrate history: the shift from jawless to jawed vertebrates. Many new features appeared in these animals\, including in the brain\, probably linked to an increasingly complex lifestyle as active predators. Yet sharks have never been classic model organisms\, as they have long been challenging to work with in the lab. As a result\, there is a major gap in our knowledge: we still do not know which brain innovations truly emerged at that time\, and we have virtually no understanding of how they arose at the molecular and cellular level. Working with catshark embryos\, we found surprising similarities with mammals in brain development\, including cell types such as basal progenitors and Cajal-Retzius cells\, long thought to be mammalian or amniote innovations. These observations\, made over only a short window of development\, suggest that key features of the developing brain arose much earlier in vertebrate history than previously assumed. With my newly established group at NeuroPSI\, we now aim to go beyond these first insights and reconstruct how the jawed vertebrate brain formed and diversified\, combining single-cell and spatial multiomics\, cross-species comparisons\, developmental biology and gene editing in the shark. In this seminar\, I will present the data that led to the establishment of the group\, as well as the main lines of our recently awarded ERC project\, EvoBrainShark.
